摘要

Abstract

[Objective] The antibacterial mechanism of lipopeptide from Bacillus natto against Staphyloccocus aureus was studied. [Method] The antibacterial mechanism of lipopeptide from Bacillus natto against Staphyloccocus aureus was studied according to the effect of lipopeptide on the growth curves,membrane permeability,protein synthesis,the structure under scanning electron microscope,and respiratory metabolism. [Result] The lipopeptide could increase membrane permeability of Staphyloccocus aureus,which led the macromolecules with ultraviolet absorption in cell to leak out of the cell. Observation under scanning electron microscope revealed the holes were formed in the cell wall of Staphyloccocus aureus,and even the cell was ruptured by lipopeptide. SDS-PAGE analysis suggested that the lipopeptide had not apparent effects on proteins expression,but had some effects on synthesized quantities of some proteins. The respiratory metabolism of Staphyloccocus aureus was inhibited by lipopeptide. [Conclusion] The lipopeptide acted on the cell membrane of Staphyloccocus aureus,which induced membrane permeability of Staphyloccocus aureus increased,or holes were formed in the cell membrane,thus the materials in the cell to leak out of it,resulted in inhibiting the growth of cell ,and killing the cell.
