Abstract
Objective:Effect of NaCl stimulation on the growth,membrane fluidity and membrane fatty acid composition of Lactobacillus bulgaricus ATCC11842 were investigated.Methods:Fluorescent probe(1,6-diphenyl-1,3,5-hexatirene,DPH)were used to analyze the fluidity(membrane polarization)of the cellular cytoplasmic membrane after stimulation of different concentrations of NaCl levels.Meanwhile,Gas chromatography(GC)was used to analyze fatty acid composition of cytoplasmic membrane under the same conditions.Results:The maiximum tolerance of L.bulgaricus was up to 1.0mol/L NaCl.Major fatty acids of this bacterium were palmitic acid(16:0)and cis-vaccenic acid(18:1)under salt stimulation(0.4mol/L NaCl).The salt concentration strongly influenced the fatty acid composition.the rate of saturation to unsaturation increased from 0.57 to 0.78 as the increasing of NaCl concentrations.Meanwhile,the growth of L.bulgaricus decreased.Conclution:When the concentration of NaCl was increased from 0.2mol/L to 0.8mol/L,the bacterial cytoplasmic membrane fluidity decreased with the fluorescence polarization increasing and the ratio of the fatty acid in cytoplasmic membrane increased.关键词
