摘要
Abstract
To prepare polyclonal antibody against heavy metal lead ion and develop an indirect competitive ELISA method. Lead was coupled to vector proteins keyhole limpet hemocyanin( KLH) and bovine serum albumin(BSA) via a bifunctional chelating agent 1TCBE respectively, Pb(D HTCBE-KLH was used as immunogen and Pb( II)-ITCBE-BSA was used as coating antigen. The anti-Pb antibody was obtained after immunized to New Zealand white rabbits and an indirect competitive ELISA method was developed. by which the determination result was compared with that by ICP-MS method. The standard curve was developed with JC50 of 3. 48 ng/mL and IC15 of 0. 32 ng/mL, the total coincidence rate of the developed ELISA method and ICP-MS method was more than 97%.关键词
