摘要

Abstract

Peanut polysaccharide was extracted from cold-pressed peanut meal by high-temperature water.The orthogonal array design was applied to optimize the process of removing starch by enzymatic hydrolysis,and peanut polysaccharide was isolated and purified by DEAE-52 cellulose ion-exchange column chromatography.Under the optimum condition,the purity of peanut polysaccharide was improved significantly after enzymatic hydrolysis using thermostable α-amylase and glucoamylase.Neutral polysaccharide PPS-1 and acidic polysaccharide PPS-2 were obtained through DEAE-52 column chromatography,and the content of these two kinds of polysaccharides was 17.79% and 66.93% respectively.UV spectral analysis indicated both PPS-1 and PPS-2 didn't contain nucleic acids,and that PPS-1 contained little protein.
