Effects of Tuna oil on Learning and Memory in Senile Mice Induced by D-galactose

Abstract
Objective: To investigate the effects of tuna oil on learning and memory in senile mice induced by D— galactose(Gal). Melhod: 72 mice were divided randomly into six groups: normal control(NC), model control(MC), positive control (PC), low dose(LD), medium dose(MD), high dose(HD). NC with normal saline [ 1 000 mg/(kg·d) ], while the others were treated with D-galactose[1 000 mg/(kg·d)) by subcutaneous injection. NC and MC were given I.g. Normal saline (10 mL/kg bw), LD,MD and HD groups were given I.g 10 mL/kg bw tuna oil respectively 20 mg/mL、 50 mg/mL and 100 mg/mL. PC were treated with piracetam [200 mg/(kg·d) ]. After six weeks, learning and memory ability in mice were evaluated by Morris water maze and Y-maze. Their brains were dissected out after Y-maze experiment for detecting the level of MDA, the activity of SOD and GSH-Px. Results: Compared with MC, the escape latency was obviously shortened, and the ratio of swimming time and distance in platform region in Morris water maze was increased, correct times were increased in Y-maze, time of electronic shock was shortened, the activity of SOD and GSH-Px was increased and the level of MDA was decreased of mice treated with tuna oil at different dose. Moreover, the high—dose group exhibited the similar effects with the piracetam-treated group. Conclusion; Tuna oil can improve learning and memory of senile mice induced by D—galactose.关键词
