Effect of Ginseng Resistant to Cylindrocarpon Root Rot Induced by Exogenous Methyl Jasmonate

Abstract

Cylindrocarpon root rot, caused by Cylindrocarpon destructons, is a severe disease on ginseng (Panax ginseng C.A. Meyer) in northeast China. Methyl jasmonate (MeJA) has been studied for its ability to induce plant defenses against pathogens. To determine the effects of MeJA on pathogen growth and disease severity, MeJA with concentrations ranging from 0 to 800 μg mL-1 was used in the experiment. The effects of MeJA on the biomass and colony diameter of C. destructans were examined in vitro. Two-year-old ginseng roots were drenched with MeJA (200μg·mL-1) and then transplanted into pots in a greenhouse. Two days following MeJA application, the roots were inoculated with conidia suspension of C. destructans (l×l0-6conidia·mL-1). Disease severity was assessed on inoculated roots, and the activation of defense responses was also measured. Results showed that MeJA had no effect on the growth of C. destructans at low concentrations (0-200μg·mL-1), but it strongly inhibited growth of the fungus at higher concentrations. The biomass and colony diameter of C. destructans were both decreased by 100% at concentration of 800μ·mL-1. The incidence rate and disease severity on MeJA-treated ginseng roots were significantly reduced compared with untreated roots. The incidence rate in MeJA-treated ginseng plants was reduced by 31.8%. The activities of phenylalanine amino-lyase (PAL), peroxidase (POD) and polyphenoloxidase (PPO) were significantly enhanced in treated roots. It was concluded that low concentrations of MeJA have no effect on C. destructans, but stimulate the activities of defense enzymes in treated ginseng roots and alleviate the damage of roots from C. destructans. Therefore, the control of Cylindrocarpon root rot by MeJA may involve the activation of the disease-related defense system.
