摘要

Abstract

As a new functional sugar substitute, Isomaltulose is perfectly for all sports, functional and wellness food and beverages. At present isomaltulose is industrially produced by a enzyme - converting process in which sucrose is transformed into isomaltulose. The main strains which can produce sucrose isomerase include Protaminobacter rubrum, Erwinia rhqpontici. However, the market growth of isomaltulose was inhibited because of the long production processes , high energy consumption and high costs of sucrose. In this study, a highly active sucrose isomerase and high isomaltulose - convertion rate Protaminobacter rubrum mutant strain was screened using nitrosoguanidine (NTG) - mutated method. The wild strain was mutated 60 minutes by 0. 2mg/mL NTG in the exponential phase, and the mutant strain was screened by Benedict reagent and HPLC. The mutant strain showed excellent genetic stability and the activity of sucrose isomerase could reach 1230U per gram of dried cell. Then the mutant strain was used in converting sucrose to isomaltulose. The maximum content of isomaltulose reached to 87. 7% in dried material. As a result, the activity of sucrose isomerase of mutant doubled and the maximum content of isomaltulose increased 7. 48% compared with the wild strain.
