Abstract

Response surface methodology was used to optimize the simultaneous alkaline extraction of proteins and polysaccharides fromUndaria pinnatifidabased on extraction efficiency. All four operating parameters including alkaline concentration, extraction time, extraction temperature and solvent-to-solid ratio on had a significant effect on the extraction yields of proteins and polysaccharides. The optimum process conditions for simultaneously increasing the yields of proteins and polysaccharides were found to be extraction at 55 ℃ for 5 h with an alkaline concentration of 2.2% and a solvent-to-solid ratio of 23:1 (mL/g). The actual yields of proteins and polysaccharides under the optimized conditions were 63.21% and 29.11%, respectively. Good agreement with the predicted values was found. Therefore, response surface methodology is a feasible way to optimize the simultaneous alkaline extraction of proteins and polysaccharides fromUndaria pinnatifida.
