Abstract

Organic solvents were used to extract proanthocyanidins from grape seed in this experiment and the extraction conditions were determined by orthogonal experiments as follows:the extraction solution pH value adjusted to 3, material liquid 1:12, 70% ethanol concentration, extraction temperature 70 ℃, extraction time 2 h, extracting 3 times. HPD100 resin was selected from eight kinds of macroporous adsorption resins by static experiment. The experiment results showed that the adsorption quantity and desorption rate of proanthocyanidins and the desorption liquid purity were high with HPD100 resin, good for the separation and purification of grape seed procyanidins. Dynamic experiment determined that the optimal concentration of sample loading was 8.0 mg/mL, pH value of 3, velocity of 3 BV/h. The desorption rate was the highest by 60%ethanol solution with the velocity of 2 BV/h. The purity of proanthocyanidins extracted by this technology was above 60%.
