海洋胶原低聚肽中抗氧化肽的分离及鉴定
Separation and identification of antioxidant peptides derived from marine collagen oligopeptides

Abstract
In order to investigate the activity and structure of antioxidant peptides derived from marine collagen oligopeptides (MCOP),MCOP was prepared by the method of two-step enzymolysis.On the basis of analysis of molecular weight distribution,the antioxidant activity of MCOP was determined using the method of DPPH radical scavenging activity as the indicator.The results showed that the major molecular weight of MCOP was less than 1000 u(90.79%),and small peptides with a molecular weight range of 132~576 u were dominated.The half-inhibition concentration (IC5o) of DPPH radical scavenging activity was approximately 8.0 mg/mL.Then,MCOP was separated and purified by reversed phase high performance liquid chromatography(RP-HPLC)and the DPPH radical scavenging activity of 11 fractions collected was assayed.The results indicated that all the fractions had the effect higher than MCOP.Finally,the identification of peptides in the 11 fractions was performed on the Q-TOF mass spectrometer and the DPPH radical scavenging activity of the 15 identified peptides was investigated.The results indicated that all the peptides exhibited DPPH radical scavenging activity and Gly-Arg (GR),ArgGlu-Arg(RER) and Ala-Pro(AP)showed stronger activities than MCOP.GR,RER and AP were potent antioxidant peptides.This study provides certain theoretical basis for the development and utilization of MCOP.关键词
