黄芪和麦冬复合多糖对TNBS诱导结肠炎大鼠的改善作用研究

Abstract
Objective:To investigate the amehorative effect of astragalus and ophiopogon japonicus polysaccharides compound on TNBS(2,4,6-trinitrobenzenesulfonic acid)induced colitis.Methods:Forty SPF sprague-dawley rats were randomly assigned into four groups according to the weight,which were negative control group,TNBS model group,sulphasalazine control group,astragalus and ophiopogon japonicus polysaccharides group.In addition to the blank control group,colitis of rats was induced by rectal instillation of TNBS.When the colitis model on rats was established,blank control group was given intragastrically with 0.9% saline,sulphasalazine control group was given intragastrically with sulfasalazine suspension(150 mg/kg),astragalus and ophiopogon japonicus polysaccharides group was given polysaccharide(300 mg/kg)for 14 days.After 14 days of drug delivery,body weight and disease activity index(DAI) were evaluated,weighing the spleen and thymus organ,conducting the gross and histopathological examination on colon,detecting the serum TNF-α,IL-2,IL-6,IL-10.Results:Compared with the blank control group,body weight,thymus weight of the TNBS model group animals were significantly reduced(p < 0.05).DAI,spleen weight,gross anatomy and tissue injury score of colon were significantly increased (p < 0.05).TNF-α,IL-2,IL-6 levels of serum were significantly increased (p < 0.05),IL-10 levels of serum was significantly decreased(p < 0.05).On rats treated with the sulfasalazine,astragalus and ophiopogon japonicus polysaccharides,all the above mentioned indexes were significantly improved (p < 0.05).Conclusion:Astragalus and ophiopogon japonicus polysaccharides has therapeutic effect on TNBS induced colitis of rats,and its therapeutic mechanism may be related to suppress the inflammatory cytokines (TNF-a,IL-2,IL-6) release and promote IL-10 release.关键词
