Abstract
A rapid and effective method was established for the determination of oleanolic acid and ursolic acid in fruit and vegetable drink by SPE-HPLC.After extracted by ethanol,the sample was concentrated and purified by amino solid phase extraction column.The separation of targeted compound was performed on a Waters RP C18 chromatographic column (4.6mm*250mm,5 μ m) using methanol/0.1% phosphoric acid solution (85 ∶ 15) as the mobile phase,and determined by PDA detector and quantified by external standard method peak area.The result showed a good linearity of oleanolic acid and ursolic acid within the concentration range of 10.0 μ g/mL ~ 100.0 μ g/ mL and the correlation coefficient is 0.999.The detection limit of oleanolic acid was 0.05 mg/kg and the quantitative limit was 0.15 mg/kg,the detection limit of ursolic acid was 0.08 mg/kg and the quantitative limit was 0.24 mg/kg.The recovery rate was 87.9% ~ 94.1%.The method has the advantages of simple pretreatment,good purifying effect,high sensitivity and fast detection rate,and is suitable for the separation and quantitative detection of oleanolic acid and ursolic acid in fruit and vegetable drink.关键词
