大肠杆菌ubiA过表达对CoQ产量的影响
Oover-expression of ubiA gene in Escherichia coil on the yield of CoQ

Abstract
The purpose of this study is to obtain a high-yield CoQ producing strain.The recombination of ubiA gene in pkk223-3 plasmid was confirmed by restriction enzyme digestion and sequencing;the strain with recombine plasmid was transformed,the changes of CoQ yield and the types of CoQ were analyzed.The fermentation process was optimized through several key factors:fermentation time,the numbers of pHBA supplements,the fermentation temperature and the amount of dissolved oxygen.The results showed that the strains with ubiA gene were successfully obtained,the yield of CoQ increased 95% and the types were unchanged;the optimized fermentation conditions were as following:replenishing pHBA 3 times,rotate speed 175 r/min,fermentation temperature 34℃ and fermentation for 15 hours,the CoQ yield achieved 2.08mg/g and increased 216%.In conclusion,it was the first time to use overexpressing ubiA gene by knocking out menA gene,CoQ production of mutant strain is greatly improved with the optimized fermentation conditions.The study goal was achieved.关键词
