Abstract

A comparative study on morphological characteristics and material digestion of Paeonia lactiflora seeds during seed germination was carried out to explore the key time for seed dormancy breaking and to reveal the mechanism of double dormancy formation and its breaking. The changes of anatomical structure, especially the structure and morphological changes of the embryo, were observed through paraffin section technique. The seed germs of the Paeonia lactiflora seeds used in stratification differentiated into cotyledons, radicles, hypocotyls and germs, namely the natural mature embryo of Paeonia lactiflora seed fully developed in the form, so there was no morphological dormancy of embryo. The morphological changes of embryos were significant at 15℃, while the structure of embryo was similar to that of non-stratification. Paeonia lactiflora seeds with stratification at 15℃ for 45 days (Paeonia lactiflora seeds were dehiscent), endosperm cell elongation and containing material consumption significantly, with the growth of radicle, the meristematic cells of hypocotyls were significantly elongated, a large number of nuclei were elongated and the cell division was vigorous, which was the key time for hypocotyl dormancy breaking. The radicle continued to be stratified at 15 ℃ after breaking through the seed coat, with the root gradual elongation in this process. When the 15 ℃ stratification of 122 days (Paeonia lactifora root length was 3-4 cm), the embryo was divided into the shape of the primary leaves, and then stratified at 4 ℃. At 4 ℃ stratification of 28 days, the endosperm cells around the embryo were disintegrated, the endosperm cells arranged loosely, the epicotyl converged on a large number of splitting cells, and the epicotyl elongated, which was the key time for epicotyl dormancy breaking. Based on the observation of the morphology and structure of the embryo of Paeonia lactiflora during the seed stratification process, the key time for epicotyl and hypocotyl dormancy breaking of the Paeonia lactiflora seeds was obtained, which could provide a accurate technique for the further study of the dormancy and germination of Paeonia lactiflora seeds in the academic and production practice.
