Purification of flavonoids from jackfruit peel with macroporous resin

Abstract

The best macroporous resin for purification jackfruit peel flavonoids was selected from five types resin by adsorption and desorption capacities.Based on single factor test,an orthogonal design was applied to optimize purification process.Analysis purification effect was evaluated by DPPH and ABTS radical scavenging assay.The results showed that NKA-9 macroporous resin was feasible for the purification.During the resin adsorption process,the optimum adsorption conditions were sample concentration 6 mg/mL,flow rate 1.5 mg/mL.The desorption conditions were elution concentration 70% (v/v) alcohol,with eluanting velocity 2.5 mL/min.The optimized purification increased the purity of the flavonoid extract from jackfruit peel to 80.15%.The IC50 of the final purified product for scavenging DPPH and ABTS free radicals were 0.0054,0.015 mg/mL,much better than 0.041,0.092 mg/mL of crude flavonoids.The results suggested that NKA-9 type macroporous resin can be used to purify the flavonoids from jackfruit peel.
