高通量测序分析食用玫瑰花瓣内生细菌多样性
Diversity of Endophytic Bacteria in Edible Rose Using High-throughput Sequencing Technology

Abstract
Objective:In order to investiage the endophytic bacterial community comparison,flower petal samples were collected from edible rose and ornamental rose.Method:The 16S rDNA-V4 region of flower petal collected from edible rose and ornamental rose were sequenced by High-throughput sequencing technology.Using Qiime and Mothur softwares,the number of sequences and operational taxonomic units (OTUs) for each sample was sorted and calculated,the species abundance and distribution,Alpha diversity index and difference times of species abundance among samples were analyzed.Results:The number of effective sequences and OTUs for each sample were 17 401/27(SM) and 11 143/ 24 (GM);The rarefaction curves showed that adequate sampling was achieved.The number of OTUs was close to saturation.The dominant species of omamental rose belongs to Escherichia,Achromobacter and Stenotrophomonas,with a percentage of 34.48%,13.79% and 13.79%,respectively.The dominant species of edible rose belongs to Faecalibacterium,Achromobacter and Ruminococcus,with a percentage of 25.53%,17.02% and 14.89%,respectively.Geothrix,Cryobacterium and Ruminococcus are endemic genus of edible rose.These species are not detected in ornamental rose.Conclusion:Significant difference were proved in diversity of endophytic bacteria between ornamental rose and edible rose.High-throughput sequencing technology provided more accurate and scientific data resources for the study of endophytic bacteria of flower petal.关键词
