摘要

Abstract

Objective:To screen and indentify a neutral and heat-tolerant β-glucosidase-producing Bacillus strain from the soil.Methods:The soil samples were treated and streaked onto the agar-plate containing esculin.After the samples were cultured at 37 ℃ for 48 h,the colonies that can produce larger dark brown hydrolysis cycle were picked and isolated.The strain with the highest β-glucosidase activity was re-screened with pNPG as the substrate,and was identified by the colony morphology in combination with the physiological and biochemical characteristics and the molecular biology technology.Results:20 strains that could produce β-glucosidase were obtained from soil samples.The strain with the highest β-glucosidase activity was obtained by re-screening,and was named ZJ1308 and identified as Bacillus cereus.Its β-glucosidase activity was up to 0.46 U/mL.Conclusion:This study lays a good foundation for the industrial application of β-glucosidase.
