Abstract
On the basis of single factor experiments,response surface analysis was used to screen the optimum conditions of the chitosan enzymatic degradation and study on the scavenging ability of DPPH radical of chitosan hydrolysates.According to the results of single factor experiments,the chitosan substrate concentration of 2g/100mL was used unchanged,and different enzyme substrate ratio,enzymatic reaction time,pH value of chitosan solution and reaction temperature were selected as independent variable,and the scavenging ability of DPPH radical of chitosan hydrolysates was selected as response value,then four factors and three levels test were designed by Box-Behnken central composite method.The results showed that the order of the four factors' effects on the scavenging ability of DPPH radical of chitosan hydrolysates was that pH value > enzymatic reaction temperature > ratio of enzyme and substrate > reaction time.The hydrolysis products' best optimization conditions of the scavenging ability of DPPH radical by response surface analysis was that the enzymatic reaction time was 64min,the E/S was 36%,the pH value of chitosan solution was pH4.6,and the reaction temperature was 54 ℃.The actual value of the scavenging ability of DPPH radical of chitosan hydrolysates was 92.60%,compared with the theoretical value model 92.36%,the error was 0.26%.Therefore,the hydrolyzate of chitosan obtained by the response surface method has better scavenging ability of DPPH radical,worthy of further research applications.关键词
