Abstract

This study has developed a reproducible surface plasmon resonance (SPR) immunosensor based on the inhibition format for the detection of highly toxic pollutant-aflatoxin B1 (AFB1) in peanut.Methods:The antigen of AFB1 (AFB1-OVA) was prepared by mixed anhydride method,and pre-coated onto the surface of SPR chip using EDC/NHS.Based on the principle of immune suppression,the antibody for AFB,at a stable concentration was simultaneously bound by the target AFB1 and the coating antigen AFB1-OVA.According to the SPR response value obtained under different AFB1 concentrations,the inhibition ratio was calculated and the accurate and sensitive analysis of AFB1 in food could be realized according to the inhibiting ratio curve.Results:The concentration of AFB1 antibody was established at 80 nmol/L.Under the optimized experimental concentrations,the detection limit (IC15) and sensitivity (IC50) for AFB1 using the proposed SPR immunosensor achieved to 0.0049 μg/L and 0.025 μg/L.In chosen peanut sample,the detection limit (IC15) and sensitivity (IC50) were 0.13 μg/kg and 0.74 μg/kg,respectively.The whole detection process could be finished within 5 min,and one immune chip could be reused more than 60 times.Conclusions:The developed SPR immunosensor has offered a simple,fast and low-cost detection strategy with high sensitivity for AFB1,which can meet the requirements of rapid,accurate and cheap analysis of high toxicity AFB1 in food samples.
