Optimal Techniques for the Mycelium Liquid Culture Conditions of Morchella importuna

Abstract

In this paper Morchella importuna'FY2'was used as a test material to obtain the optimal technology of Morhella liquid culture using single-factor test and orthogonal test L9 (34).The results showed that the best growth temperature was 25℃for the mycelium of Morchella importuna, the best growth initial pH was 7. The mycelium dry weight could reach 4.0 g?L-1 when inoculating amounts were 5% and 10%. Therefore the best inoculating amount was 5%. The mycelium of Morchella importuna could grow in 7 carbon sources, the best carbon source is glucose. The mycelium of Morchella importuna can grow in 6 nitrogen sources, the best nitrogen source is soymeal. The effects of temperature and initial pH on the mycelium of Morchella importuna were very significant. The effects of carbon sources on the mycelium of Morchella importuna were significant. However, the effects of nitrogen sources on the mycelium of Morchella importuna were not significant. The optimal mycelium liquid culture conditions were temperature being 20℃, initial pH 7.0, glucose concentration 2.0%, soymeal concentration 0.15%.
