蔗糖异构酶PalI包埋和交联固定化方法比较
Comparison of Immobilization Methods of Sucrose Isomerase PalI with Embedding and Crosslinking

Abstract
The immobilization methods using sodium alginate as the embedding medium and flutaralde-h≥de as the crosslinker was used to improve the c≥clic utilization and stabilit≥ of immobilized sucrose isomerase PalI from Klebsiella sp. LX3. The results showed that the highest enz≥me activit≥was obatained when the concentrations of sodium alginate and CaCl2 were 1. 5% and 2% . Meanwhile, the optimum temperature and pH value of embedding enz≥me were 40 ℃ and 6. 0. The optimum condition of the crosslinking method was the concentrations of ammonium sulfate and flutaraldeh≥de were 90% and 2. 5% . Meanwhile, the optimum temperature and pH value of crosslinking enz≥me were 50℃ and 5. 0. Through the comparison of stabilit≥, the conclusion was that the enz≥me activit≥ of both methods was better than that of free enz≥me. The residual enz≥me activit≥ of immobilized enz≥me with flutaraldeh≥de and sodium alginate kept in 95% and 60% , while that of the free enz≥me was onl≥30% when enz≥mes were stored at 4℃ for 20 da≥s. The crosslinking immobilization enz≥me was superior to the sodium algi-nate immobilized enz≥me, and its residual enz≥me activit≥was still 80% after repeated use for 12 times.关键词
