Tuina Promotes Muscle Injury Repair in Exercised-induced Muscle Injury Rats by Regulating Hippo Signaling Pathway

目的:探究推拿对过度使用性骨骼肌损伤大鼠的修复作用和Hippo信号通路及生肌调节因子的影响.方法:将24只雄性SD大鼠随机分为对照组(C组)、模型组(M组),模型+推拿组(T组)及模型+假推组(G组),每组6只.M、T、G进行4周大负荷运动建立EIMD模型;T组运动后6h施加推拿按压手法,G组同期予以毛刷轻抚.透射电镜观察肌纤维超微结构变化,HE染色评估肌纤维横截面积CSA,实时定量RT-PCR检测MST1、YAP、TAZ、TEAD2的mRNA表达量,Western Blot检测PAX7、MYOD、MYOG蛋白表达量,结果:(1)行为学结果表示模型组和假推组大鼠相对抓力比值增加,CSA下降(P<0.05),推拿可进一步增加四肢相对抓力比值,增加CSA(P<0.05;透射电镜显示模型组和假推组肌纤维肌节肿胀、Z线断裂、细胞排列紊乱,推拿一定程度上促进了肌纤维修复;模型组和假推组YAP、TAZ、TEAD2 mRNA表达降低,MST1 mRNA表达升高,推拿可升高YAP、TAZ、TEAD2 mRNA表达,降低MST1 mRNA(P<0.05);模型组和假推组Pax7、MYOD蛋白表达降低,MYOG蛋白表达升高,推拿可提升PAX7、MYOD蛋白表达,降低MYOG蛋白表达.结论:推拿能有效促进过度使用性肌损伤的良性修复,其机制可能与调节hippo信号通路和生肌调节因子有关.

Objective:To explore the repair effect of tuina on overuse-induced skeletal muscle injury in rats and the effects of Hippo signaling path-way and myogenic regulatory factors.Methods:Twenty-four male SD rats were randomly divided into control group(C group),model group(M group),model + tuina group(T group)and model + sham tuina group(G group),with 6 rats in each group.EIMD model was established by 4-week heavy load exercise in group M,T,G;Group T was treated with tuian by pressing 6 hours after exercise,and Group G was treated with brush at the same time.Ultrastructural changes of muscle fibers were observed by transmission electron microscope,cross-sectional area of muscle fibers was evaluated by HE staining,mRNA expression of MST1,YAP,TAZ,TEAD2 was detected by real-time quantitative RT-PCR,and expression of PAX7,MYOD,MYOG protein was detected by Western Blot.Results:(1)Behavioral results showed that the ratio of grasping force/body mass of limbs in-creased and CSA decreased in model group and sham push group(P<0.05).Tuina could further increase the ratio of grasping force/body mass of limbs and increase CSA(P<0.05);Transmission electron microscope showed that muscle fiber sarcomere swelling,Z-line rupture and cell arrange-ment disorder in model group and sham tuina group.Tuina promoted muscle fiber repair to a certain extent;The expression of YAP,TAZ,TEAD2 mRNA decreased and the expression of MST1 mRNA increased in model group and sham tuina group,but the expression of YAP,TAZ,TEAD2 mRNA increased and the expression of MST1 mRNA decreased(P<0.05);The expression of PAX7,MYOD protein decreased and the expression of MYOG protein increased in model group and sham tuina group.Tuina increased the expression of PAX7,MYOD protein and decreased the expres-sion of MYOG protein.Conclusion:tuina can effectively promote the benign repair of overused muscle injury,and the mechanism may be related to the regulation of hippo signaling pathway and myogenic regulatory factors.
