Abstract
An efficient and rapid method for the determination of flavonoids in Ginkgo biloba L.leaves was established,and the content differences of flavonoids in Ginkgo biloba L.leaves from three different places were analyzed.The purification process of flavonoids from Ginkgo biloba L.leaves was explored and optimized.The results showed that the liquid chromatography conditions were as follows:ZORBAX AQ-C18 column(250 mm×4.6 mm,5 μm)injection volume:10 μL;flow rate:1 mL/min;detection time:17 min;mobile phase:phase A was methanol,phase B was 0.4%phosphoric acid aqueous solution;gradient elution:(1~5 min:58%A-42%B;5~17 min:50%A-50%B);column temperature:40℃;The UV wavelength of the detector was 360 nm.Under this condition,quercetin,isorhamnetin and kaempferol in the flavonoids of Ginkgo biloba L.leaves had high peak separation and good peak shape,which could be used for qualitative and quantitative detection.The purification results showed that the content of Ginkgo biloba L.flavonoids in the powder was up to 73.5%after vacuum drying under the conditions of the eluent with the concentration of 39.9 mg/mL,the volume of 15mL,the elution rate of 1 BV/h and 70%ethanol.Therefore,the established method for the detection of flavonoids in Ginkgo biloba L.leaves was stable and reliable.The optimized purification process was conducive to improve the purity of flavonoids from Ginkgo biloba L.leaves.The study provides reference and basis for further research and application of flavonoids from Ginkgo biloba L.leaves.关键词
