摘要

Abstract

MiR-140-3p is a miRNA widely expressed in gonadal tissues of different species.qRT-PCR was used to detect the expression difference between male and female of miR-140-3p in gonads of Parab-gurnus dabryanus,and its biological function was analyzed by predicting related target genes.The results showed that miR-140-3p was expressed in both testis and ovaries,and the expression level of miR-140-3p in testis was significantly higher than that in ovary(P<0.01).KEGG pathway analysis showed that target genes CPEB4 and GDF9 were involved in oocyte maturation and differentiation,and were enriched in pro-gesterone mediated oocyte maturation pathway,oocyte meiosis pathway and ovarian steroid generation pathway,respectively.These pathways were related to gonad development and reproduction.qRT-PCR re-sults showed that the expression of CPEB4 and GDF9 in ovary was significantly higher than that in testis(P<0.01),and the expression level was significantly negatively correlated with expression of miR-140-3p,and these results suggest that miR-140-3p has a potential target relationship with CPEB4 and GDF9.This study provides basic information for future exploration of the role of miR-140-3p in gonad development and reproduction of fish.
