miR-140-3p是在不同物种性腺组织中广泛表达的一种miRNA.本研究利用qRT-PCR方法检测miR-140-3p在大鳞副泥鳅性腺中的雌雄表达差异并预测其生物学功能.结果显示:miR-140-3p在大鳞副泥鳅雌雄鱼的性腺组织中均有表达,且精巢中的表达量显著高于卵巢(P<0.01);KEGG pathway分析表明,靶基因CPEB4和GDF9参与卵母细胞成熟分化过程,富集在与性腺发育、繁殖功能相关的孕激素介导卵母细胞成熟通路、卵母细胞减数分裂通路、卵巢类固醇生成通路.荧光定量结果显示CPEB4和GDF9在卵巢中表达量显著高于精巢(P<0.01),且表达量与miR-140-3p的表达呈显著负相关,说明miR-140-3p和CPEB4、GDF9具有潜在的靶标关系.这为进一步探索miR-140-3p在鱼类性腺发育和繁殖过程中的作用奠定了基础.

MiR-140-3p is a miRNA widely expressed in gonadal tissues of different species.qRT-PCR was used to detect the expression difference between male and female of miR-140-3p in gonads of Parab-gurnus dabryanus,and its biological function was analyzed by predicting related target genes.The results showed that miR-140-3p was expressed in both testis and ovaries,and the expression level of miR-140-3p in testis was significantly higher than that in ovary(P<0.01).KEGG pathway analysis showed that target genes CPEB4 and GDF9 were involved in oocyte maturation and differentiation,and were enriched in pro-gesterone mediated oocyte maturation pathway,oocyte meiosis pathway and ovarian steroid generation pathway,respectively.These pathways were related to gonad development and reproduction.qRT-PCR re-sults showed that the expression of CPEB4 and GDF9 in ovary was significantly higher than that in testis(P<0.01),and the expression level was significantly negatively correlated with expression of miR-140-3p,and these results suggest that miR-140-3p has a potential target relationship with CPEB4 and GDF9.This study provides basic information for future exploration of the role of miR-140-3p in gonad development and reproduction of fish.
