响应面法提取优化破壁灵芝孢子粉多糖及其分级醇沉组分的抗氧化活性研究
Extraction Optimization of Broken Ganoderma lucidum Spore Powder Polysaccharides by Response Surface Methodology and Antioxidant Activity of Ethanol Fractional Precipitation

Abstract
[Objective]The optimum extraction process of Broken Ganoderma lucidum Spore Powder polysaccharide(BGP)and antioxidant activity of ethanol fractional precipitation were investigated.[Method]The Broken Ganoderma lucidum Spore Powder was as material,the internal boiling method was used to explore the extracting temperature,extraction time,and the ratio of material and liquid to the yield of BGP.Response surface analysis was used to optimize the extraction parameters of BGP.Different concentrations of ethanol were used to precipitation BGP,the antioxidant activity of polysaccharides was determined by measuring the scavenging ability of DPPH free radical and hydroxyl free radical.[Result]According to the analysis of the regression model,the best extraction parameters are the extraction temperature 83 ℃,the extraction time 7 min,and the ratio of material and liquid 1:21.At these experiment parameters,the obtained BGP content was 9.10%.Different concentrations of ethanol were used to precipitation BGP,and five BGP were obtained.The results showed that when the concentration of ethanol was 90%,the BGP content was the highest.Antioxidant experiments showed that each ethanol precipitation BGP had a certain antioxidant activity.The antioxidant of 90%-BGP was slightly stronger than other BGP,and as the polysaccharide concentration increased,the antioxidant resistance of each ethanol precipitation BGP had a certain increase.[Conclu-sion]Extract using the optimal extraction parameters,the different ethanol fractional precipitation BGP with certain antioxidant activity.关键词
