秋水仙素对不同葡萄多倍体诱导的影响OA
Effect of Colchicine on Polyploid Induction in Explants of Different Grape Variaties
采用不同浓度(100、250、500 mg·L-1)秋水仙素延时处理(1、2、3 d后再培养6个月)砧木葡萄'5BB'的不定芽、胚性愈伤组织和体细胞胚以选出适于后续试验的外植体类型.数据显示,选用不定芽作为外植体较为合适,秋水仙素处理后成苗时间用时最短,为5周,而胚性愈伤和体细胞胚成苗用时分别为22周、20周.进而沿用相同秋水仙素浓度梯度处理'红宝石无核''金星无核''阳光玫瑰''白奥林'和'藤稔'葡萄的不定芽,并检测秋水仙素对'红宝石无核'再生植株的染色体加倍效果及对植株性状的影响.结果发现,当秋水仙素浓度越高、处理时间越长时,不定芽存活率越低,秋水仙素处理显著延缓植株再生速度;且处理后的各品种间亦显示出不同差异,处理后1~2个月,'金星无核'和'白奥林'存活率快速下降,而'红宝石无核''阳光玫瑰'和'藤稔'葡萄存活率下降明显较为缓慢.同时,秋水仙素处理后再生植株的染色体出现不同水平的加倍,并表现为外观上的差异,即具有112条染色体的多倍体葡萄植株叶片大小、叶绿素相对含量、节间长度、根系质量显著高于二倍体对照植株;虽然染色体数目增加的植株茎粗与对照相比差异并不显著,但植株叶片的气孔大小高于对照,气孔密度却显著降低.
In this study,different concentrations(100、250、500 mg·L-1)of colchicine were used for delayed treatment(1,2,3 days and then cultured for six months)of different types of rootstock'5BB'explants and different table grape varieties.Adventitious buds,embryogenic callus and somatic embryos were used to screen the type of explant suitable for subsequent experiments.The data showed that adventitious buds were more suitable as explants.The seedling growth time after colchicine treatment was the shortest(5 weeks),while the seedling growth times of embryogenic callus and somatic embryos were 22 weeks and 20 weeks,respectively.Then,the same colchicine concentration gradient was used to treat the adventitious buds of'Ruby Seedless''Venus Seedless''Shine Muscat''White Olin'and'Fujiminori'grapevines,the chromosomal doubling effect of colchicine on'Ruby Seedless'regenerated plants and its effect on the characteristics of grapevine plants were tested.The results showed that the survival rate of explants would decline as the colchicine concentration went higher and treatment duration lasted longer.Meanwhile,colchicine treatment significantly delayed the plant regeneration rate,the adventitious buds were the appropriate explant for a great deal of grape chromosome doubling research as they could be obtained easily and regenerate into plantlets within a short time.After colchicine treatment and grown for 1-2 months,the survival rate of'Venus Seedless'and'White Olympia'decreased rapidly,while the survival rate of'Ruby Seedless''Shine Muscat'and'Fujiminori'varieties declined more slowly.The chromosome numbers of plantlets developed from adventitious buds treated with colchicine were improved at different levels,the chromosome doubling grape plantlets containing 112 chromosomes distinctly had larger leaf,longer internode length,higher chlorophyll relative content and higher weight of the root system,although the stem diameter of plants with increased chromosome number was not significantly different from that of the control,the stomatal size of leaves was higher than that of the control,but the stomatal density was significantly decreased.
