目的:观察骨宝胶囊对糖皮质激素性骨质疏松大鼠骨密度、骨微结构的影响.方法:将40只雄性Wistar大鼠随机分为空白组、模型组、给药组(骨宝胶囊)、对照组(钙尔奇D),每组大鼠数量为10只.模型组、给药组、对照组通过给予地塞米松磷酸钠注射液(2.5 mg/kg)注射,空白组予以等体积0.9%氯化钠注射液(2.5 mL/kg)注射,每周2次(周一、周四),持续8周.造模8周后,给药组灌胃骨宝胶囊0.4 g/(kg·d),对照组灌胃钙尔奇D 60 mg/(kg·d),空白组、模型组灌胃等体积生理盐水,每天1次.灌胃12周后给予2%戊巴比妥钠腹腔注射并处死大鼠.获取离心血清,测定血清钙、血清磷、碱性磷酸酶水平;取大鼠左侧股骨通过Micro-CT行计算机断层扫描并三维重建感兴趣区域图像,观察分析各组骨骼情况;采用苏木精-伊红染色法观察大鼠右侧股骨标本组织病理变化.结果:血清生化指标显示,给药组、对照组血清钙水平明显高于模型组(P<0.05);Micro-CT图像和三维重建图形显示,模型组骨小梁结构稀疏,经骨宝胶囊、钙尔奇D药物治疗后,BMD、BV/TV、Tb.N明显增加,Tb.Sp下降显著(P<0.05),CT与三维图像表明给药组对骨密度、骨微结构的改善效果整体略优于对照组;苏木精-伊红染色结果显示,模型组骨小梁稀疏、断裂、分离,而给药组和对照组的骨小梁数量增加,结构较密.结论:骨宝胶囊能通过影响糖皮质激素大鼠的骨密度、骨微结构、骨代谢的方式改善骨质疏松.

Objective:To observe the effect of Gubao Capsule on bone mineral density and bone microstructure in rats with glucocorticoid-induced osteoporosis.Methods:Forty male Wistar rats were randomly divided into blank group,model group,drug administration group(Gubao Capsule)and control group(Caltrate D),with 10 rats in each group.The model group,drug administration group and control group were injected with dexamethasone sodium phosphate(2.5 mg/kg),and the blank group was injected with 0.9%sodium chloride(2.5 mL/kg)twice a week(Monday and Thursday)for 8 weeks.After successful modeling,the drug administration group was gavaged Gubao Capsules[0.4 g/(kg·d)],the control group was gavaged Caltrate D 60 mg/(kg·d),and the blank group and model group were gavaged the same volume of normal saline,once a day.Intraperitoneal injection of 2%pentobarbital sodium was administered after 12 weeks of gastric lavage and the rats were euthanized.Centrifuge serum was obtained and serum calcium,serum phosphorus and alkaline phosphatase levels were determined.Computed tomography(CT)was done for the left femur of rats by Micro-CT and the image of the region of interest was reconstructed in 3D to observe and analyze the skeletal conditions of each group.Hematoxylin-eosin staining was used to observe the histopathological changes of right femur specimens.Results:The serum biochemical indexes showed that the serum calcium level in the administration group and the control group was significantly higher than that in the model group(P<0.05).Micro-CT images and 3D reconstruction graphics showed that the bone trabecular structure was sparse in the model group.After the treatment with Gubao Capsule and Caltrate D,BMD,BV/TV,Tb.N increased significantly,and Tb.Sp decreased significantly(P<0.05).CT and 3D images showed that the improvement of bone mineral density and bone microstructure in the administration group was slightly superior to that in the control group.The results of hematoxylin-eosin staining showed that the bone trabeculae were sparse,fractured and separated in the model group,while the number of bone trabeculae increased and the structure was dense in the drug administration group and the control group.Conclusion:Gubao Capsule can improve osteoporosis by affecting bone mineral density,bone microstructure and bone metabolism in glucocorticoid rats.
