山东省临沭县鸭疫里默氏杆菌分离鉴定及生物特性分析OA
Isolation,Identification and Biological Characteristics Analysis of Riemerella anatipestifer in Linshu County,Shandong Province
为了解山东省临沭县鸭疫里默氏杆菌(RA)血清型、耐药性和致病性,2023 年 5-6 月在 8 家养殖场无菌采集疑似感染RA的病死鸭鹅心血、脑和肝脏组织病料,利用细菌分离培养、生化试验和PCR方法对分离菌进行鉴定,用玻片凝集试验鉴定分离菌血清型,并对分离菌进行药敏性和致病性分析.结果显示:共有12株分离菌的菌落特性、镜检、生化特点和 16S rRNA基因序列符合RA特征;有 7 株RA为血清 1 型,2 株为血清2型,3株为血清11型;所有菌株均对丁胺卡那、庆大霉素、新霉素和阿莫西林耐药,对头孢他啶、头孢曲松、头孢氨苄敏感;用3 种血清型RA攻毒均可导致雏鸭发病死亡,且临床症状、病理变化与RA感染典型特点相同.结果表明:临沭县RA感染比较普遍,主要流行 3 种血清型,其中血清 1 型为优势血清型;RA对多种抗菌药表现出耐药性,致病性较强.建议加强定期病原学监测和耐药性分析,结合本地菌株生物学特性选择疫苗和抗菌药防治,以提高RA感染防控效果.
In order to investigate the serotype,drug resistance and pathogenicity of Riemerella anatipestifer(RA)in Linshu County,Shandong Province,heart blood,brain and liver tissues of dead ducks and geese suspected of being infected with RA were aseptically collected on 8 farms from May to June 2023.After bacterial isolation and culture,the isolates were identified by biochemical test and PCR assay,and then their serotypes were determined by slide agglutination test,followed by analysis on their drug sensitivity and pathogenicity.The results showed that there were 12 isolates whose colony characteristics,microscopic examination results,biochemical characteristics and 16S rRNA gene sequence were consistent with RA;7,2 and 3 strains of RA fell into serotype 1,2 and 11,respectively;all the strains were resistant to amikacin,gentamicin,neomycin and amoxicillin,but sensitive to ceftazidime,ceftriaxone and cefalexin.The three serotypes of RA,after challenge test,could make ducklings die of infection,with same clinical symptoms and pathological changes as the typical characteristics of RA infection.In conclusion,RA infection was widely available in Linshu County,mainly including three serotypes,in which serotype 1 was dominant;RA was resistant to various antibiotics with strong pathogenicity.Regular pathogen monitoring and drug resistance analysis should be strengthened,and targeted vaccines and antibiotics should be selected based on the biological characteristics of local strains to improve the effectiveness of RA prevention and control.
