推拿按揉法对慢性坐骨神经挤压伤大鼠脊髓背角星形胶质细胞和神经元的影响
Effects of Tuina on Astrocytes and Neurons in Spinal Dorsal Horn of Rats with Chronic Sciatic Nerve Crush Injury

Abstract
Objective:To observe the effect of tuina on astrocytes and neurons in rats with chronic constriction injury(CCI),and to explore the poten-tial mechanism of action of tuina in the treatment of neuropathic pain.Methods:Forty SD rats were randomly divided into 5 groups(n=8):control group,model group,tuina group,astrocyte inhibitor group and astrocyte inhibitor + tuina group.All groups except the control group were established CCI model.From the 4th day after modeling,the tuina group received pressing-kneading intervention for 10 minutes/time/day,and the astrocyte in-hibitor group received intrathecal injection of fluorocitrate(FCA)10ul/rat/day.The astrocyte inhibitor + tuina group was treated with intrathecal in-jection of FCA and pressing and knead Weizhong points for 14 days.The expression of glial fibrillary acidic protein(GFAP)protein in the spinal dor-sal horn was detected by immunofluorescence.The changes in the morphology,arrangement and number of neurons in the spinal dorsal horn were ob-served by Ni staining.The correlation between GFAP expression level and the number of surviving neurons was analyzed.Results:Compared with the control group,the mechanical pain threshold of the model group decreased significantly(P<0.01);After tuina intervention,the threshold of the tuina group gradually increased compared with the model group,and there was a significant difference after 14 days of cumulative treatment(P<0.01).From the 7th day of FCA injection,the pain threshold of the astrocyte inhibitor group and the astrocyte inhibitor + tuina group increased sig-nificantly(P<0.01).Compared with the control group,the expression of GFAP in the spinal dorsal horn of the model group was significantly in-creased(P<0.01),the morphology of neurons was irregular,loosely arranged,and the number of neurons was reduced(P<0.01).After tuina treat-ment,GFAP expression was down-regulated(P<0.01),the morphology of neurons tended to be regular,the arrangement tended to be neat,and the number of neurons increased(P<0.01).After FCA injection,GFAP expression levels in the astrocyte inhibitor group and the astrocyte inhibitor+tuina group were significantly lower than those in the model group(P<0.01).Compared with the model group,the morphology and arrangement of neu-rons in the model group were more regular and the number was increased(P<0.01).Correlation analysis showed that the mean fluorescence intensity of GFAP was negatively correlated with the number of surviving neurons(r=-0.4649,P<0.05).Conclusion:Tuina can alleviate the pain behavior of CCI rats and reduce the damage of spinal dorsal horn neurons,and its mechanism may be related to the inhibition of astrocyte activation.关键词
