目的:基于网络药理学方法探讨理中汤在过敏性鼻炎治疗中的潜在靶点及其作用机制,为临床应用及今后研究提供方向与借鉴.方法:应用多个数据平台剖析理中汤治疗过敏性鼻炎的机制,从中药系统药理学数据库与分析平台中检索筛选理中汤的有效成分和相关靶点,采用全球蛋白质资源(Uniprot)数据库进行标准化靶点.利用基因卡片(GeneCards)、在线人类孟德尔遗传(OMIM)、比较毒理基因组学(CTD)、治疗靶点(ITD)等数据库查找过敏性鼻炎的相关基因.通过韦恩图获得两者交集治疗靶点并将其导入STRING数据库中分析,并建立蛋白相互作用(PPI)网络.最后对核心关键靶点进行基因本位(GO)功能富集和京都基因与基因组百科全书(KEGG)信号通路分析.结果:理中汤治疗过敏性鼻炎的关键化学成分可能为槲皮素、山柰酚、β-谷甾醇、α-香树素和柚皮素等;核心靶点包括肿瘤坏死因子(TNF)、丝/苏氨酸蛋白激酶(AKT1)、白细胞介素-6(IL-6)、肿瘤抑制基因P53(TP53)及IL-1β等;理中汤在过敏性鼻炎治疗中主要涉及IL-17信号通路、晚期糖基化终产物及其受体(AGE-RAGE)信号通路、脂质和动脉粥样硬化信号通路、TNF信号通路、Toll样受体信号通路及Th17细胞分化通路等.结论:本研究初步揭示了理中汤治疗过敏性鼻炎的多成分、多靶点、多通路的作用机制,为理中汤在临床应用及实验研究等方面提供了参考.

Objective:To analyze the potential therapeutic targets and mechanism of Lizhong decoction in the treatment of allergic rhinitis based on network pharmacology,so as to provide direction and reference for clinical application and later research.Methods:Mechanism of Lizhong decoction in the treatment of allergic rhinitis was analyzed by using multi-data platforms,the active components and drug targets of Lizhong decoction were retrieved and screened in Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Targets were standardized with Uniprot database.GeneCards,Online Mendelian Inheritance in Man(OMIM),Comparative Toxicogenomics Database(CTD),Therapeutic Target Database(TTD)databases were retrieved to search and obtain the related genes of allergic rhinitis.Intersection therapeutic targets were obtained by Venn diagram and imported into STRING database for analysis,and the protein-protein interaction(PPI)network was established.The core key targets were enriched by gene ontology(GO)function and Kyoto Encyclopedia of genes and genomes(KEGG)signaling pathway analysis.Results:The key chemical components of Lizhong decoction in the treatment of allergic rhinitis were quercetin,kaempferol,β-sitosterol,α-amyrin,naringenin.The core targets were tumor necrosis factor(TNF),serine/threonine protein kinase(AKT1),interleukin-6(IL-6),tumor suppressor gene P53(TP53)and interleukin-1β(IL-1β).Lizhong decoction signaling pathway in the treatment of allergic rhinitis mainly included IL-17 signaling pathway,advanced glycation end products-receptor for advanced glycation end products(AGE-RAGE)signaling pathway,lipid and atherosclerosis signaling pathway,TNF signaling pathway,Toll-like receptor signaling pathway,and Th17 cell differentiation pathway.Conclusion:This study preliminarily reveals the multi-component,multi-target and multi-channel mechanism of Lizhong decoction in the treatment of allergic rhinitis,and provided reference for the clinical application and experimental research of Lizhong decoction.
