目的:从网络药理学角度探讨赖氨肌醇维生素B12口服液治疗小儿厌食症的机制.方法:通过查阅药品说明书,明确赖氨肌醇维生素B12口服液的成分,并通过Swiss Target Prediction网站预测成分的相关靶点,与小儿厌食症靶点进行交集映射,获得其治疗小儿厌食症的作用靶点.利用蛋白相互作用(PPI)网络筛选核心靶点,并进行分子对接验证.基于共有靶点,采用功能注释生物信息学芯片进行分析,得到赖氨肌醇维生素B12口服液治疗小儿厌食症的关键通路.结果:赖氨肌醇维生素B12口服液中的主要成分为盐酸赖氨酸、肌醇和维生素B12,共预测出40个靶基因,其中有13个靶基因与小儿厌食症的关系密切,主要涉及谷氨酸受体信号通路、神经活性配体-受体相互作用等相关分子环节.结论:赖氨肌醇维生素B12口服液可能发挥类似于谷氨酸的作用,通过激活突触后膜的谷氨酸受体,提高神经兴奋性,从而改善患儿的厌食症状,体现了多成分、多靶点协同起效的作用特点.

Objective:To explore the mechanism of lysine,inosite and vitamin B12 oral solution in the treatment of anorexia in children based on network pharmacology.Methods:By referring to drug instructions,the components of lysine,inosite and vitamin B12 oral solution were excavated,the correlation targets of components were predicted through Swiss Target Prediction website,and the intersection mapping was made with the targets of anorexia,so as to obtain the therapeutic targets of anorexia in children.Protein-protein interaction(PPI)was used to screen core targets and molecular docking verification was carried out.Based on common targets,functional annotation bioinformatics chip was used for analysis,and the key pathway of lysine,inosite and vitamin B12 oral solution in the treatment of anorexia in children was obtained.Results:Lysine hydrochloride,inositol and vitamin B12 were the main components in lysine,inosite and vitamin B12 oral solution.A total of 40 target genes were predicted,among which 13 target genes were closely related to anorexism,mainly including glutamate receptor signaling pathway and neuroactive ligand receptor interaction.Conclusion:Lysine,inosite and vitamin B12 oral solution may play a similar role to glutamate,and improve anorexia in children by activating glutamate receptors in the postsynaptic membrane,which reflects the synergistic effect of multiple components and multiple targets.
