探究春砂仁多糖(AVP)对高脂饮食引起的小鼠脂代谢异常和氧化应激的改善作用.试验选取120 只 6 周龄健康的C57BL/6雄性小鼠,随机分为:基础日粮组(CK)、高脂日粮组(HFD)、HFD+AVP低剂量组(HFD+L-AVP)、HFD+AVP中剂量组(HFD+M-AVP)和HFD+AVP高剂量组(HFD+H-AVP),试验共 8 周.结果表明:日粮中添加AVP,降低了高脂饲喂小鼠血清中AST、ALT、LDL-C、TC和TG的水平,提高了HDL-C水平.提高了小鼠肝脏和肠道中SOD、CAT和GSH-Px的活性,显著降低了肝脏和肠道中MDA水平.降低了高脂饮食小鼠SREBP1 和FAS的mRNA表达水平,提高了ATGL的mRNA表达水平,显著抑制小鼠的脂肪生成.AVP的添加显著提高了高脂饮食小鼠肠道中Nrf2、HO-1 mRNA和蛋白的表达水平,也提高其肠道抗氧化能力.综上所述,春砂仁多糖可缓解高脂饮食导致的小鼠氧化应激和脂代谢异常.

To explore the effect of Amomum villosum polysaccharide(AVP)on abnormal lipid metabolism and oxidative stress in mice induced by high-fat diet for 8 weeks.A total of 120 6-week-old healthy C57BL/6 male mice were randomly divided into control check group(CK),high-fat diet group(HFD),HFD+AVP low-dose group(HFD+L-AVP),HFD+AVP medium-dose group(HFD+M-AVP)and HFD+AVP high-dose group(HFD+H-AVP).The results showed that,in the group fed with AVP in the diet,the mice serum aspartate transaminase(AST),alanine aminotransferase(ALT),low-density lipoprotein(LDL-C)levels,triglyceride(TG)and total cholesterol(TC)were all significantly decreased.The high-density lipoprotein(HDL-C)in the serum of mice increased significantly.The activities of superoxide dismutase(SOD),peroxidase(CAT)and glutathione peroxidase(GSH-Px)in the liver and intestine of mice with high-fat diet were increased,and the level of malondialdehyde(MDA)was significantly reduced.AVP decreased mRNA expression level of cholesterol regulatory element binding protein 1(SREBP1)and fatty acid synthase(FAS)in high-fat diet mice,and mRNA expression level of triglyceride hydrolase(ATGL)in high-fat diet mice was increased.AVP significantly increased expression level of nuclear factor E2 related factor 2(Nrf2),heme oxygenase 1(HO-1)mRNA and protein in the intestine of high-fat diet mice and improved the intestinal antioxidant capacity.To sum up,Amomum villosum polysaccharide alleviated oxidative stress and abnormal lipid metabolism in mice caused by high-fat diet.
