白花蛇舌草诱导活性氧类物质抑制慢性髓系白血病K562细胞增殖
Mechanism of hedyotis diffusa willd inhibiting the proliferation of K562 cells in chronic mye-loid leukemia by inducing reactive oxygen species

Abstract
Aim To explore the effects of hedyotis diffusa willd(HDW)on the proliferation of K562 cells in chronic mye-loid leukemia(CML)by inducing reactive oxygen species(ROS).Methods ROS content in K562 cells treated with HDW was detected by flow cytometry.K562 cells were divided into control group,HDW group,N-acetyl-L-cysteine group(NAC group)and HDW+NAC group.ROS content,cells activity,morphology,proliferation and distribution of cells cycle in different groups were measured.The expressions of cells cycle-related proteins,phosphorylated phosphatidylinositol 3-kinase(p-PI3K)and serine threo-nine kinase(p-Akt)proteins were detected by Western blotting.Results HDW could increase ROS content in K562 cells(P<0.05).In control group and NAC group,there were a large number of trophochrome cells,and there was no significant change in cells morphology.In HDW group and HDW+NAC group,trophochrome cells were reduced,staining cells were increased,and there was chromatin aggregation and nuclear membrane disintegration.Compared with control group,ROS content,proportion of cells in G1 phase and expressions of p21 and p53 proteins were increased,while proportion of cells in S phase and expressions of cyclin D1,cyclin-dependent kinase 4(CDK4),p-PI3K and P-Akt proteins were decreased in HDW group(P<0.05).NAC intervention could partially reverse the effect of HDW(P<0.05).Conclusion HDW can decrease proliferation ability of K562 cells by inducing the increase of ROS,which is related to cell cycle arrest and regulating activity of PI3K/Akt pathways.关键词
