1800 MHz电磁辐射暴露对大鼠外周血单个核细胞代谢组的影响OACSTPCD
Effect of 1800 MHz electromagnetic radiation exposure on metabolome of peripheral blood mononuclear cells in rats
目的 分析电磁辐射暴露对大鼠外周血单个核细胞(PBMCs)代谢物变化的影响.方法 将24只SD大鼠随机分成电磁辐射暴露组(EMR组)和假暴露组(Sham组).EMR组大鼠置于中心频率1 800 MHz、平均功率密度284.6 V/m的电磁混响箱内,每天暴露10 min,连续暴露7 d;Sham组辐照功率为0 W,其他条件及操作与EMR组相同.7 d后,收集大鼠外周血并分离PBMCs进行液相色谱/质谱联用(LC-MS/MS)非靶向代谢组学分析.使用偏最小二乘法-判别分析(PLS-DA)进行主成分分析,单变量分析筛选差异代谢物,再联合mzCloud和ChemSpider数据库对检测到的代谢物进行鉴定,构建代谢通路以分析电磁辐射暴露对大鼠PBMCs代谢组的影响.结果 Sham组与EMR组SD大鼠PBMCs正负离子模式下共检测到差异代谢物2 017种,正离子模式下1 611种,其中847种上调,764种下调;负离子模式下406种,其中220种上调,186种下调.正负离子模式下检测到的差异代谢物包括氨基酸、肽类、有机酸、胆汁酸等.KEGG富集分析显示,正负离子模式下差异代谢物涉及的代谢通路共64个,包括氨基酸代谢,脂类代谢,细胞生长、增殖及死亡,氧化应激,炎症反应等.结论 模拟职业环境电磁辐射暴露可以对SD大鼠PBMCs代谢组产生明显影响,其差异代谢产物有望成为电磁辐射暴露的生物标志物.
Objective To analyze the effect of electromagnetic radiation exposure on the metabolite change of peripheral blood mononuclear cells(PBMCs)in rats.Methods A total of 24 SD rats were randomly divided into the electromagnetic radiation group(EMR group)and the Sham group.The rats in the EMR group were placed in an electromagnetic reverberation box with a center frequency of 1 800 MHz and an average power density of 284.6 V/m,and were exposed for 10 minutes every day,and continuously for 7 days;the rats in the Sham group were given an irradiation power of 0 W,and other conditions and operations were the same as those of the EMR group.After exposure for 7 days,peripheral blood of rats was collected and PBMCs were isolated for liquid chromatography-tandem mass spectrometry(LC-MS/MS)untargeted metabolomics analysis.Principal component analysis was conducted using partial least squares-discriminant analysis(PLA-DA),univariate analysis was used to screen differential metabolites,and then combined with mzCloud and ChemSpider databases to identify the detected metabolites,and a metabolic pathway was constructed to analyze the effects of electromagnetic radiation exposure on the metabolome of PBMCs in rats.Results A total of 2 017 differential metabolites were detected from the PBMCs under the positive and negative ion modes of the SD rats in the EMR group and the Sham group,among which 1 611 metabolites were obtained under the positive ion mode,with 847 up-regulated metabolites and 764 down-regulated metabolites,and 406 metabolites were obtained under the negative ion mode,with 220 up-regulated metabolites,and 186 down-regulated metabolites.Differential metabolites detected in the positive and negative ion modes included amino acids,peptides,organic acids,bile acids,and so on.KEGG enrichment analysis showed that there were 64 metabolic pathways involved in the differential metabolites under the positive and negative ion modes,including amino acid metabolism,lipid metabolism,cell growth,cell proliferation and cell death,oxidative stress,and inflammatory response.Conclusion Simulated occupational environment exposure to electromagnetic radiation has a significant effect on the metabolome of PBMCs in SD rats,and its differential metabolites are expected to be biomarkers of electromagnetic exposure.
