Analysis of intestinal flora early colonizing in Hu-sheep of different genders
摘要
Abstract
[Objective]In order to provide basic data for later study of the relationship between early intestinal flora and the occurrence of body health or diseases,the early intestinal flora colonizing in Hu-sheep of different genders was analyzed.[Method]The feces of 48 45-day-old Hu-sheep(31 ewes and 17 rams)were collected for extracting genomic DNA,and high-throughput sequencing was per-formed to screen effective sequences and operational taxonomic units(OTUs).Then,the intestinal mi-crobial diversity of Hu-sheep of different genders was analyzed,and the specific differences in intesti-nal flora at phyla and genus levels were identified by linear discriminant analysis effect size(LEfSe),non-metric multidimensional scaling analysis(NMDS)and short-term attention/memory priority(STAMP)models.[Result]By high throughput sequencing technology,2 014 OTUs were obtained from ewes,while 1 747 OTUs were obtained from rams,and 1 637 OTUs were obtained from both ewes and rams.A total of 15 phyla,25 classes,62 orders,1 18 families and 317 genera were identified in the early colonized intestinal flora in different gender Hu-sheep.There were significant differences in intestinal flora at phylum level and genus level among different gender Hu-sheep in early period.At the phylum level,Firmicutes and Bacteroidota were the dominant phyla,the highest relative abundance was Firmicutes(49.68%)in ewes,while the Bacteroidea(47.34%)was the highest in rams,and the proportion of Elusmicrobiota in ewes was significantly higher than that in rams.At the genus level,there were fifteen kinds of genera with gender difference,most of which were concentrated in Firmi-cutes.Among them,the proportion of Succinivibrio and Anaerostipes which were colonized in rams in-testinal tract was significantly higher than that in ewes.The proportion of Monoglobus,Eubacterium_nodatum and Elusimicrobium were significantly lower than that in ewes.[Conclusion]The diversity and richness of intestinal flora in ewes Hu-sheep were higher than that of rams,and there were gender differences at phylum and genus level.关键词
