目的:探讨藏药七十味珍珠丸(Tibetan medicine Ranasampel,RNSP)对帕金森病(Parkinson dis-ease,PD)模型大鼠认知能力的影响.方法:采用6-羟基多巴胺(6-hydroxydopamine,6-OHDA)右侧纹状体单点注射法制作PD大鼠模型,建模成功后将动物随机分为模型组,西药组(美多芭50 mg/kg),RNSP低、中、高剂量组(90、180、360 mg/kg)及正常组,共6组,每组8只.造模次日各组大鼠灌胃给药,每日1次,连续4周,模型组和正常组大鼠予以等体积生理盐水.给药当天及第7、14、21、28天分别称量动物体重;RNSP给药4周后进行阿扑吗啡诱导的大鼠旋转实验及Morris水迷宫实验;免疫组化法观察大鼠中脑黑质致密部及腹侧被盖区酪氨酸羟化酶(tyrosine hy-droxylase,TH)阳性神经元数目;酶联免疫吸附法测定大鼠纹状体氧化应激指标丙二醛,还原型谷胱甘肽及超氧化物歧化酶水平;高效液相色谱电化学法检测大鼠海马多巴胺、5-羟色胺以及去甲肾上腺素含量;Western blot 检测大鼠黑质P38、p-P38、JNK、p-JNK、ERK和p-ERK蛋白表达.结果:RNSP给药4周后各组大鼠之间体重无显著差异;与正常组相比,PD模型组大鼠旋转频率显著增加,逃避潜伏期显著延长,穿台次数及目标象限停留时间显著减少(P<0.05或P<0.01);黑质致密部及腹侧被盖区TH阳性神经元数目显著减少;纹状体还原型谷胱甘肽及超氧化物歧化酶含量显著降低,丙二醛含量显著升高(P<0.05);海马多巴胺、5-羟色胺以及去甲肾上腺素含量显著降低;黑质p-JNK/JNK、p-P38/P38比值显著升高(P<0.05).与PD模型组相比,RNSP各组大鼠旋转频率显著降低,逃避潜伏期显著缩短,穿台次数及目标象限停留时间显著增加(P<0.05);TH阳性神经元数目、还原型谷胱甘肽,超氧化物歧化酶、多巴胺含量显著升高,丙二醛含量及p-JNK/JNK、p-P38/P38比值显著降低(P<0.05).中、高剂量组去甲肾上腺素含量及低、高剂量组5-羟色胺含量显著升高,中、高剂量组p-ERK/ERK比值显著降低(P<0.05).结论:RNSP可改善PD动物认知能力,其机制可能与抑制P38/JNK/ERK信号通路,减轻中脑氧化应激反应,减少DA神经元丢失,提高海马单胺类神经递质有关.

AIM:To investigate the effect of Tibetan medicine Ranasampel(RNSP)on the cognitive ability of Parkinson disease(PD)model rats.METHODS:The PD rats were established by 6-hydroxydopamine(6-OHDA)single-point injection in right striatum.The animals were randomly divided into PD model group,Medopar group(Medopar 50 mg/kg),RNSP low-dose,medium-dose,high-dose group(90,180,360 mg/kg)and normal group,with 8 rats in each group.The rats in each group were intragastrically administered once a day for 4 weeks after modeling,while the PD mod-el group and normal group received equal volume of saline.The rats were weighed on first day of RNSP administration,then on the 7th,14th,21th and 28th days.After 4 weeks of RNSP administration,apomorphine induced Rotation experi-ment and the Morris water maze experiment were performed.The number of tyrosine hydroxylase(TH)positive neurons in the substantia nigra densities(SNc)and ventral tegmental area(VTA)of the midbrain was observed by immunohistochem-istry.The levels of malonaldehyde,reduced glutathione and superoxide dismutase in the striatum of rats were measured by enzyme-linked immunosorbent assay.The levels of dopamine,5-hydroxytryptamine and noradrenaline in hippocampus of rats were detected by high performance liquid chromatography electrochemical method.The protein expressions of P38,p-P38,JNK,p-JNK,ERK and p-ERK in the substantia nigra of rats were detected by Western blot.RESULTS:After 4 weeks of RNSP administration,there was no significant different in body weight among the groups.Compared with normal group,the rotation frequency of the PD model group was increased significantly,the escape latency was significantly pro-longed,the number of table crossing times and the target-zone duration time were significantly decreased(P<0.05 or P<0.01).The number of TH positive neurons were significantly reduced in SNc and VTA;The levels of reduce glutathione and superoxide dismutase in the striatum were significantly decreased,and the level of malondialdehyde was significantly increased(P<0.05).The levels of dopamine,5-hydroxytryptamine and norepinephrine in the hippocampus were de-creased;The ratios of p-JNK/JNK and p-P38/P38 in the substantia nigra were significantly increased(P<0.05).Com-pared with PD model group,the rotation frequency of the RNSP groups was significantly decreased,the escape latency was significantly shortened,and the number of table crossing times and target-zone duration time were significantly increased(P<0.05).The number of TH-positive neurons,and the levels of reduced glutathione,superoxide dismutase,and dopa-mine were significantly increased,while the level of malondialdehyde and the ratios of p-JNK/JNK and p-P38/P38 were sig-nificantly decreased(P<0.05).The noradrenaline level in the medium and high dose groups and the 5-hydroxytryptamine level in the low and high dose groups were significantly increased,and the ratio of p-ERK/ERK in the medium and high dose groups were significantly decreased(P<0.05).CONCLUSION:RNSP can improve the cognitive ability of PD mod-el rats,and the mechanism may be related to inhibiting the P38/JNK/ERK signaling pathway,reducing the oxidative stress response in the midbrain,reducing the loss of DA neurons,and improving the monoamine neurotransmitters in the hippo-campus.
