陕西部分地区长角血蜱的鉴定及其基因多态性OA北大核心CSTPCD

Identification and Gene Polymorphism of Haemaphysalis longicornis in Some Areas of Shaanxi Province

为比较陕西省渭南市和商洛市长角血蜱的核糖体基因差异,探讨长角血蜱的分子鉴定方法,先通过体式显微镜观察蜱的形态结构进行形态学鉴定确认.再对长角血蜱的18S、ITS1、5.8S、ITS2和28S核糖体序列进行测定,比对与边缘革蜱等常见蜱种的基因的同源性并构建系统发育树,分析基因多态性,筛选有效标记.结果显示,核糖体5.8S序列不能区分不同种属的蜱虫,18S、28S、ITS1和ITS2序列可区分不同种属的蜱虫.虽然ITS1和ITS2多态性高于18S,但是18S不仅可以将国内和国外长角血蜱区分开,还可以区分国内不同地域的长角血蜱.18S序列区分不同长角血蜱地域株能力最强,可作为长角血蜱分子鉴定的有效标记.

This experiment was conducted to compare the differences in ribosome genes of Haemaphysalis longicornis,which collected from Weinan city and Shangluo city,Shaanxi province,and explore appropriate molecular markers to provide a foundation for Haemaphysalis longicornis identification.Morphological structures of ticks were observed by stereomicroscope;the ribosome 18S,ITS1,5.8S,ITS2 and 28S sequences of Haemaphysalis longicornis were determined by molecular biology techniques,the homology of genes was compared,and the phylogenetic tree was constructed to analyze the gene polymorphism.The results showed that 5.8S sequence of ribosome could not distinguish ticks from different species,while 18S,28S,ITS1 and ITS2 sequences could distinguish ticks from different species.Though 18S and ITS1 sequences could distinguish Haemaphysalis longicornis from China and overseas,Haemaphysalis longicornis from different regions in China could be distinguished by the 18S sequence.By comparing the ribosome gene fragments of Haemaphysalis longicornis in Shaanxi province,the phylogenetic relationship showed that the 18S sequence has the strongest ability to distinguish different region strains of Haemaphysalis longicornis and could be used as an effective tool for the classification and molecular identification of Haemaphysalis longicornis.
