目的:基于卵巢细胞外基质研究黄连对多囊卵巢综合征(PCOS)小鼠的治疗作用.方法:选取6 只C57BL/6 小鼠作为对照组(Control 组).选取 24 只 C57BL/6 小鼠皮下注射 21d 脱氢表雄酮(DHEA)构建PCOS模型,其中 6 只作为模型组(PCOS 组),其余 18 只分别使用 NaCl(PCOS + NaCl组)、200 g·kg-1·d-1黄连活性成分(PCOS +RC200 组)以及400 g·kg-1·d-1黄连活性成分(PCOS + RC400 组)进行灌胃处理28 d.Western blot法检测各组小鼠卵巢细胞外基质重塑相关蛋白MMP-2、COL-I及LOX-1 表达水平.使用KGN细胞过表达和敲低YTHDF2,分为PC3.1 组、PC3.1-YTHDF2组、NC组和SiYTHDF2 组.Western blot法检测MMP-2 蛋白表达水平.结果:与Control组相比,PCOS组卵巢中MMP-2、COL-I及LOX-1 蛋白表达水平均显著升高(P<0.05).与PCOS +NaCl组相比,PCOS +RC200 组和PCOS + RC400 组卵巢中MMP-2、COL-I及LOX-1 蛋白表达水平均显著降低(P<0.05).与PC3.1 组相比,PC3.1-YTHDF2 组MMP-2、COL-I及LOX-1 的表达水平显著升高(P<0.05).与NC组相比,siYTHDF2 组MMP-2 和COL-I的表达水平显著下降(P<0.05).结论:黄连可能通过调节MMP-2、COL-I及LOX-1 改善PCOS卵巢细胞外基质重塑状态,从而影响排卵.YTHDF2 可影响KGN细胞中MMP-2、COL-I及LOX-1 的表达水平,可能为黄连治疗PCOS的靶点之一.

Objective:To explore the therapeutic effect of Rhizoma Coptidis on polycystic ovary syndrome(PCOS)mice based on ovarian extracellular matrix.Methods:Six C57BL/6 mice were selected as control group.Twenty-four C57BL/6 mice were subcutaneously injected with 21 d dehydroepiandrosterone(DHEA)to construct PCOS model.Six of them were in the model group(PCOS group),and the other 18 animals were treated with NaCl(PCOS + NaCl group),200 g·kg-1·d-1 active ingredient of Rhizoma Coptidis(PCOS + RC200 group)and 400 g·kg-1·d-1 active ingredient of Rhizoma Coptidis(PCOS + RC400 group)by gavage for 28 days,respectively.The expression levels of extracellular matrix remodeling related proteins MMP-2,COL-I and LOX-1 were detected by Western blot.Using KGN cells overetpressing and knocking down YTHDF2,mice were divided into PC3 1group,PC3 1-YTHDF2 group,and SIYTHDF2 group.Western blot was used to detect MMP-2 protein expression level.Results:Compared to control group,the levels of MMP-2,COL-I,and LOX-1 in the ovaries of PCOS group significantly increased(P<0.05).Compared to PCOS + NaCl group,the levels of MMP-2,COL-I,and LOX-1 in the ovaries of PCOS + RC200 group and PCOS + RC400 group significantly decreased(P<0.05).Compared to PC3.1 group,MMP-2,COL-I and LOX-1 levels significantly increased in PC3.1-YTHDF2 group(P<0.05).Compared to NC group,MMP-2 and COL-I levels significantly decreased in siYTHDF2 group(P<0.05).Conclusion:Rhizoma Coptidis may improve the extracellular matrix remodeling of PCOS ovaries by regulating MMP-2,COL-I,and LOX-1,thereby affecting ovulation.YTHDF2 can affect the expression levels of MMP-2,COL-I,and LOX-1 in KGN cells,and may be one of the targets for Rhizoma Coptidis treatment of PCOS.
