为了评价牛源金黄色葡萄球菌J581株和无乳链球菌A20株荚膜多糖蛋白结合疫苗的免疫原性,对制备的3批荚膜多糖进行了多糖、蛋白质及核酸含量检测,同时分别用纯化的J581和A20株荚膜多糖与破伤风类毒素偶联制备了质量浓度分别为5、10、15 μg/mL的6种荚膜多糖蛋白结合疫苗及质量浓度各为10 μg/mL的J581和A20株荚膜多糖蛋白结合混合疫苗,用这几种疫苗对小鼠进行免疫,采用间接ELISA方法及IgG试剂盒对免疫前后不同时间段的小鼠血清进行抗J581和A20株荚膜多糖的特异性抗体水平及IgG抗体效价检测.结果表明,制备的3批纯化荚膜多糖中,J581多糖平均质量浓度为49.78 mg/L,A20多糖平均质量浓度为55.76 g/L,2种多糖中蛋白和核酸含量均低于1%,符合疫苗生产要求;小鼠免疫试验结果表明,6种不同多糖含量的荚膜多糖蛋白结合疫苗和荚膜多糖混合疫苗均能刺激机体产生一定水平的特异性抗体效价及IgG抗体,且在第28天抗体水平达到最高,其中荚膜多糖蛋白混合疫苗组中J581和A20抗体效价及特异性IgG水平均显著高于6种单疫苗组(P<0.05).本研究为研制金黄色葡萄球菌和无乳链球菌荚膜多糖蛋白结合疫苗奠定了基础.

To evaluate the immunogenicity of vaccines made from capsular polysaccharide-protein conjugate from bovine Streptococcus agalactiae A20 strain and Staphylococcus aureus J581 strain,three batches of capsular polysaccharides were tested for polysaccharides,proteins,and nucleic acids.In addition,purified capsular polysaccharides of S.aureus J581 strain and S.agalactiae A20 strain were combined with tetanus toxoid to prepare six types of conjugate vaccines with polysaccharide-protein concentrations of 5 μg/mL,10 μg/mL,and 15 μg/mL,as well as a mixture vaccine with polysaccharide-protein mass concentrations of 10 μg/mL from J581 and A20 capsules.In the end,mice were immunized separately with these varying vaccine types.The indirect ELISA method and IgG antibody kit were used to determine specific antibody levels and antibody titers against J581 and A20 capsular polysaccharides in mouse serum at different time periods before and after vaccination.Among the three batches of purified capsular polysaccharides prepared,the average mass concentration of J581 polysaccharide was 49.78 mg/L,and the average mass concentration of A20 polysaccharide was 55.76 g/L.These two polysaccharides contained less than 1%protein and nucleic acid,which was consistent with vaccine production requirements.The results of the mouse immune experiment showed that six types of capsular polysaccharide-protein conjugate vaccines and capsular polysaccharide mixed vaccines with different polysaccharide contents can stimulate the body to produce a certain level of specific antibody titers and IgG antibodies,which reach a specific level on the 28th day.J581 and A20 antibody titers and specific IgG levels were significantly higher in the mixed vaccine group than those in the 6 single vaccine groups(P<0.05).The study lays the foundation for developing vaccines against S.aureus and S.agalactiae containing capsular polysaccharide-protein conjugates.
