WRKY是植物特有的转录因子,在生长发育和逆境胁迫响应过程中起着重要作用.为了明确BoiWRKY25在霜霉病抗性反应中的作用,本研究以青花菜为材料,利用PCR法克隆该基因,在序列分析和表达分析的基础上,利用过量表达研究其功能.结果表明,BoiWRKY25的基因组全长为1 395 bp,有4个内含子.BoiWRKY25的编码区全长为1 086 bp,编码361个氨基酸,分子量为40.4 kDa,等电点为6.98.BoiWRKY25的平均亲水性指数为-0.857,是一种亲水蛋白.BoiWRKY25具2个WRKY结构域,每个结构域由1个WRKYGQK七肽和1个C2H2型锌指结构组成,锌指结构可分别用C-X4-C-X22-H-X1-H和C-X4-C-X23-H-X1-H表示.基因表达结果表明,BoiWRKY25的表达受霜霉菌的诱导,表达量在72 h时达到最大值,为对照的4.6倍;过量表达植株对霜霉病的抗性显著降低,同时病程相关蛋白基因BoiPR1的表达量降低,表明BoiWRKY25在霜霉病抗性反应中起负调控作用.转录因子基因BoiWRKY25的克隆与表达分析,为后续开展青花菜霜霉病抗性机理研究及分子育种奠定了基础.

WRKYs are plant-specific transcription factors,play important roles in growth,development,and stress responses.To elucidate the function of BoiWRKY25 in resistance response to downy mildew,in this study,broccoli was used as the material to isolate the gene by PCR method,the gene's function was investigated through gene over-expression based on sequence analysis and expression analysis.The results indicated that BoiWRKY25 had a total length of 1 395 bp,with four introns.The complete coding region of BoiWRKY25 was 1086 bp,encoding a protein of 361 amino acids with a molecular weight of 40.4 kDa and an isoelectric point of 6.98.The grand average hydropathicity of BoiWRKY25 was-0.857,and it was determined as a hydrophilic protein.BoiWRKY25 featured two WRKY domains,each comprising a WRKYGQK heptapeptide and a C2H2-type zinc finger structure.The two zinc finger structures could be represented as C-X4-C-X22-H-X1-H and C-X4-C-X23-H-X1-H,respectively.Expression analysis revealed that BoiWRKY25 was induced by downy mildew,and the highest relative level of expression was observed at 72 h post-inoculation,with a fold change of 4.6 as compared to the control.Over-expression of BoiWRKY25 in broccoli lines reduced the resistance to downy mildew,and the expression levels of the pathogenesis-related protein gene BoiPR1 were significantly down-regulated,indicating BoiWRKY25 plays a negative regulatory role in the resistance response against downy mildew.Isolation and expression analysis of BoiWRKY25 shed light on the mechanism of downy mildew resistance and molecular breeding in broccoli for future research.
