Abstract

The nutrients composition of Chlorella growth factor(CGF)was determined by phenol-sulfuric acid method,Coomassie brilliant blue method and ultraviolet spectrophotometry.The protective effect of CGF on ultraviolet-injured human skin fibroblasts(HSF)was determined by MTT method and the expression of collagen-related factors were determined by enzyme-linked immunosorbent(ELISA),respectively.The results showed that nucleic acids accounted for 84.40%±0.73%of CGF.Pretreatment with Chlorella Growth Factor significantly reduced the proliferation inhibition of HSF cell by UVA.Compared with the model group,treatment with a 50 μg/mL concentration of CGF increased HSF cell viability by 75.05%,showing a dose-dependent relationship within 5~50μg/mL of CGF.ELISA results revealed that Chlorella growth factor could increase the expression of tissue inhibitors of metalloproteinase-1(TIMP-1)and type I collagen(COL-I)in HSF cells and reduce the expression of matrix metalloproteinase-1(MMP-1).Chlorella growth factor is an excellent exogenous nucleotide nutrient that can effectively alleviate the damage caused by UVA exposure,possibly through promoting collagen synthesis.
