磁性微球是一种多用途复合材料,通常以纤维素、壳聚糖、二氧化硅以及聚苯乙烯等生物兼容性好的材料为包覆载体,通过表面的活性基团(—OH、—COOH、—NH2和—SH等)负载荧光分子、抗体和核酸等,可以特异性识别癌细胞、病毒和遗传物质.借助其自身优异的磁响应特性,磁性微球可实现快速分离和富集,在免疫分析、细胞分离以及环境检测等领域具有广阔的应用前景;磁性微球还可与微流控芯片等结合,构建高通量检测平台.本文介绍了近5年来磁性微球在循环肿瘤细胞检测、病毒抗原抗体和核酸检测等前沿分析领域中的应用进展,并讨论了以磁性微球以及以其为载体开发的检测分析试剂盒的商业化前景.

Magnetic beads (MB),composed of magnetic nanoparticles and biocompatible materials including cellulose,chitosan,silica and polystyrene,are ideal platforms for specific detection of cancer cells,virus and nucleic acids when loaded with antibodies,fluorescent and conjugation molecules. With the ease of separations using magnets,magnetic particles show tremendous advantages over other platforms and thus arising potentials in circulating tumor cell (CTC),virus (such as SARS-CoV-2) antibody and antigen detections,and nucleic acid extractions and purifications. In addition,when combined with microfluidic devices,magnetic particles can be used to develop detection platforms with high throughputs,as well as portable and point-of-care apparatus. In accompany with their huge potentials in all kinds of biomedical and clinical scenarios,magnetic particles based medical devices have been commercialized such as antigen test kits that have been commonly used during the pandemic. In this review,the progress in detailed antibody conjugation practice used in each application scenario based on MB platform was summarized and the commercialization of analytical devices combined with microfluidics and others was discussed.
