海红米糠多糖的提取工艺、结构表征及抑制HepG2细胞增殖研究
Study on extraction technology,structure characterization,and inhibition of HepG2 cell proliferation of sea red rice bran polysaccharide

Abstract
The objective of this study was to study the extraction technology,structural characterization of sea red rice bran polysac-charide(SRBP)and its inhibition of HepG2 cell proliferation.Taking sea red rice bran as raw material,the effects of different extraction methods on the yield of SRBP were investigated.The optimal extraction process of SRBP was optimized by the response surface method on the basis of a single factor.The chemical composition was determined by phenol-sulfuric acid method and m-hydroxy-biphenyl colorimetry.The molecular weight and monosaccharide composition were determined by liquid chromatography,and the structure was characterized by ultraviolet-visible,fourier transform infrared spectroscopy,and scanning electron microscopy.The effect of SRBP on the growth of HepG2 cells was determined by methyl thiazolye tetrazolium(MTT)assay.The experimental results showed that the optimal extraction conditions of SRBP were an enzymolysis time of 61 min,solid-liquid ratio of 1∶33(g∶mL),and enzymolysis temperature of 59℃.Results showed that SRBP was a high-purity mannose-pyrane-type heteropolysaccharide,which did not contain protein and nucleic acid substances,and had a rough surface and sheet structure.MTT experiment showed that the survival rate of HepG2 cells treated with SRBP at 800 μg/mL for 24 h was only(9.77±0.81)%,indicating that SRBP had the ability to inhibit the proliferation of HepG2 cells.In conclusion,the ex-tracted SRBP has certain anti-tumor activity,which could provide a theoretical basis and data reference for dietary intervention in liver cancer.关键词
