为了探究黄曲霉毒素B1(AFB1)呈现双向免疫毒性作用的时间和剂量,通过剂量梯度试验和时间梯度试验,考察不同剂量、时间的AFB1暴露对小鼠体重、各脏器指数、剖检变化和病理特征的影响,明确AFB1暴露对小鼠的双向免疫毒性作用.剂量梯度试验选取雄性昆明鼠30只,随机分为6组(每组5只),每天灌服0,10,20,40,80,160 μg·kg-1 AFB1(按体重计),试验持续21 d;时间梯度试验采用固定剂量的AFB1每日对小鼠进行腹腔注射,分别持续0,7,14,21,28,35 d.试验结果表明:与对照组(0 μg·kg-1)相比,在较高浓度(80,160 μg·kg-1)和较长时间(21 d)的AFB1作用下,小鼠的体重显著降低(p<0.01),脾脏指数、胸腺指数和血清中TNF-α表达水平呈现先升高而后降低的趋势;相反,IL-10在较高浓度(80,160 μg·kg-1)和较长时间(21 d)AFB1作用下则表现显著升高趋势,脾脏和胸腺的损伤程度也表现相似的结果.综上可知,低浓度或短时间AFB1暴露可能诱发炎症,而高浓度或长时间AFB1暴露可能导致免疫抑制,证实了不同时间和剂量AFB1暴露可以诱导双向免疫毒性作用,研究结果将为AFB1污染的防控和致病机理的深入研究提供科学依据.

In order to study the time points and dose of aflatoxin B1(AFB1)showing"bidirectional immunotoxicity",the effects of AFB1 exposure at different doses and times on body weight,organ index,autopsy changes and pathological characteristics of mice were investigated by dose gradient test and time gradient test,and the bidirectional immunotoxicity effect of AFB1 exposure on mice was clarified.In the dose gradient test,30 male Kunming mice were randomly divided into 6 groups(5 mice in each group),and daily given 0,10,20,40,80,160 μg·kg-1 of AFB1(by weight),the test lasted for 21 days.In the time gradient test,mice were intraperitoneally injected daily with a fixed dose of AFB1 for 0,7,14,21,28,35 days,respectively.The results showed that mice exposed to higher concentrations(80,160 μg·kg-1)and a longer period(21 days)of AFB1 had significantly lower weight gain(p<0.01),and the expression levels of TNF-α,spleen index and thymus index increased first and decreased then than those exposed to the control group(0 μg·kg-1),while IL-10 showed a significant increase in higher concentrations(80,160 μg·kg-1)and longer period(21 days)of AFB1 exposure,and the degree of damage to the spleen and thymus also showed similar results.In conclusion,low concentrations or short periods of AFB1 exposure may induce inflammation,while high concentrations or prolonged AFB1 exposure may lead to immunosuppression,which confirms that different times and doses of AFB1 exposure can induce bidirectional immunotoxic effects,and the results of this study will provide a scientific basis for the prevention and control of AFB1 pollution and the in-depth study of the pathogenic mechanism.
