摘要

Abstract

Volvariella volvacea VG39 is a mutant strain previously obtained through mutagenesis of V.volvacea V23 by atmospheric pressure room temperature plasma.It exhibits a certain level of tolerance to low-temperature stress,but the molecular characteristics in response to low-temperature stress remain unclear.Both VG39 and V23(control)were incubated under low temperature(4 ℃)and then measured for contents of polysaccharides,lysine,soluble protein and total free amino acids in the mycelia,detected for infrared spectrum,and analyzed for transcriptome.The results showed that VG39 contained higher levels of polysaccharides and lysine than V23 before 4 ℃ treatment.After 4 ℃ treatment for 8 h,both strains contained significantly lower levels of polysaccharides,lysine,soluble protein,and total amino acids compared with those without 4 ℃ treatment.The number of the infrared absorption peaks in VG39 was also significantly decreased after 4 ℃ treatment for 8 h.Compared with before 4 ℃ treatment,there were 882 differentially expressed genes(DEGs)up-regulated in VG39 after 4 ℃ treatment for 8 h.The number of DEGs in ribosomes(2 up-regulated genes and 12 down-regulated genes)was the greatest,followed by the number of DEGs in the glycine,serine and threonine metabolic pathway(8 up-regulated genes).These results provided a reference for understanding the underlying mechanism of VG39 in response to low-temperature stress.
