Physicochemical Properties of Naematelia aurantialba Crude Polysaccharides Extracted with Different Solvents and Their Effects on Cell Migration

Abstract
The effects of different extraction solvents on the chemical composition,structural characteristics,and rheological properties of Naematelia aurantialba crude polysaccharide extracts were studied.The crude polysacchride extracts were also studied for their toxicity to HaCaT cells and effect on HaCaT migration.Different solvents,distilled water,0.1 mol·L-1 NaOH solution and 0.1 mol·L-1 HC1 solution,were used to extract fruiting bodies of N.aurantialba to generate three crude polysaccharide extracts NAP-WE(extracted by water),NAP-A1E(extracted by NaOH)and NAP-AcE(extracted by HC1),respectively.NAP-WE,NAP-A1E and NAP-AcE were then determined for total sugar content by the phenol-sulfuric acid method,reducing sugar content by the dinitrosalicylic acid method,uronic acid content by the m-hydroxy-biphenyl method,molecular weight and distribution by high performance size exclusion chromatography-multiangle laser light scattering-refractive index(HPSEC-MALLS-RI),characteristic groups by Fourier transform infrared spectrometry,monosaccharide composition by ion chromatography,apparent viscosity and modulus by a rheometer,and cytotoxicity and cell mobility in vitro.The results showed that the yield of NAP-WE was higher than the other two crude polysaccharide extracts,the contents of polysaccharide and uronic acid in NAP-A1E were higher than those in the other two crude polysaccharide extracts,and the protein content of NAP-AcE was higher than that of the other two crude polysaccharide extracts.NAP-WE had a large Mw of 2.462×106 g·mol-1,and its polydispersion coefficient(Mw/Mn)was 1.515.The Mw and polydispersion coefficient of NAP-A1E were 1.187×106 g·mol-1 and 1.530,respectively.NAP-AcE had the lowest Mwof 4.545×105 g·mol-1 among the three crude polysaccharide extracts,and its polydispersion coefficient was 2.501.All three kinds of crude polysaccharide extracts contained the characteristic polysaccharide absorption peaks of hydroxyl,carboxyl,α and β glucosidic bonds,and were composed of xylose,mannose,glucuronic acid and a small amount of glucose.Compared with NAP-WE and NAP-AcE,NAP-A1E contained higher levels of glucose and glucuronic acid in molar percentage,but a lower molar percentage of xylose.The molar percentage of mannose in NAP-WE was lower than that in NAP-A1E and NAP-AcE.When the mass concentration of the crude polysaccharide extracts was 40-60 mg·mL-1,the apparent viscosity was NAP-WE>NAP-AlE>NAP-AcE.NAP-WE and NAP-A1E formed an entangled network structure when the mass concentration was 30-60 mg·mL-1and 60 mg·mL-1,respectively.On the other hand,NAP-AcE remained in a fluid state at the mass concentration range of 20-60 mg·mL-1.When the mass concentration was 50-800 pg·mL-1,all three crude polysaccharide extracts had no obvious cytotoxicity to HaCaT cells.They promoted HaCaT cell migration at the mass concentration range of 200-800 pg·mL-1.These results provided a reference for research on N.aurantialba polysaccharides.关键词
