摘要

Abstract

To address the limitations of traditional high-fructose syrup production,such as low catalytic efficiency of conventional glucose isomerases,this study focused on the heterologous expression and optimization of a novel glucose isomerase,Pd-xylA,derived from Pantoea dispersa isolated from kiwifruit roots.The Pd-xylA gene was cloned and expressed in Escherichia coli BL21(DE3),followed by systematic optimization of induction conditions including temperature and IPTG(isopropyl-β-D-thiogalactopyranoside)concentration.The results demonstrated that the optimal induction conditions were 28℃and 0.1 mmol/L IPTG,under which Pd-xylA achieved the highest soluble expression level.This study provided theoretical support for the development of enzyme resources with enhanced catalytic activity and industrial applicability,thereby advancing the biotechnological production of high-fructose syrup.
