Optimization of Yunnan Coffea arabica pericarp pigment purification process using macroporous resin and its antioxidant activities

Abstract

In order to study the optimal extraction and purification process and antioxidant activity of the pigments from the pericarp of Yunnan Coffea arabica,the optimal extraction and purification process of the pigments was determined by response surface method optimization test and dynamic adsorption-desorption test,and the scavenging ability of the crude pigments and the purified pigments against DPPH·,ABTS+·and·OH was investigated.The results showed that the optimal extraction process for the pigment of the pericarp of Yunnan Coffea arabica was:ethanol volume fraction of 52%,ultrasonic power of 241 W,ultrasonic time of 48 min,and liquid-to-material ratio of 55 mL/g,and the extraction rate reached 74.47%under these conditions.The optimal purification conditions using X-5 microporous resin were:up-sample concentration of 6 mg/mL,up-sample flow rate of 2 mL/min,desorption solution ethanol volume fraction of 60%,desorption flow rate of 3 mL/min,and the pigment purification efficiency was 55.38%under these conditions.The IC50 values for the crude pigment in scavenging DPPH·,ABTS+·and·OH were(0.1788±0.0124),(0.4084±0.0115),and(0.2492±0.0152)mg/mL,respectively;The IC50 values for the purified pigment in scavenging DPPH·,ABTS+·and·OH were(0.0774±0.0007),(0.1220±0.0047)and(0.0963±0.0114)mg/mL,respectively.The pigments extracted from Yunnan Coffea arabica pericarp had good antioxidant properties,and the antioxidant capacity of the purified pigments was significantly enhanced.The results provide a theoretical basis for the further processing of Yunnan Coffea arabica pericarp pigments.
