Abstract

Hemp defatted protein was used as the raw material,with the inhibition rate of Dipeptidyl Peptidase Ⅳ(DPP-Ⅳ)as the evaluation criteria,neutral protease was employed for the hydrolysis to prepare hemp peptides.Based on single factor experiments,the enzymatic hydrolysis process of hemp peptides was optimized using response surface methodology.The results showed that the optimal enzymatic hydrolysis process of hemp defatted protein was:a material to liquid ratio of 1:8 g/mL,enzyme addition of 5.7%,and enzymatic hydrolysis time of 6 hours.Under these conditions,the inhibition rate of the DPP-Ⅳ by the hydrolysis product reached 38.24%.The hydrolysis product was fractionated into four components of different molecular weights by ultrafiltration,among which the peptides with a molecular weight of less than 1 kDa exhibited the strongest hypoglycemic activity,with DPP-Ⅳ inhibition rates of 41.06%,α-amylase inhibition rates of 64.79%,and α-glucosidase inhibition rates of 48.51%.This study lays the foundation for the further separation and purification of hypoglycemic peptides from hemp defatted protein and demonstrates certain research application value in the development of therapeutic special medical foods for diabetes.
