响应面法优化草酸铵提取银杏叶多糖工艺及抗氧化活性研究
Optimization of the extraction process of polysaccharide from Ginkgo biloba L.leaf using ammonium oxalate via response surface methodology and its antioxidant activity

Abstract
The effects of various extraction methods on properties of polysaccharide from Ginkgo biloba L.leaf were investigated.The response surface methodology(RSM)was employed to optimize the extraction process of Ginkgo biloba L.leaf polysaccharides using ammonium oxalate.Titration and infrared spectroscopy were employed to characterize the physicochemical properties of the polysaccharides extracted by the three methods.Antioxidant activity of polysaccharides obtained from these extraction methods was assessed through DPPH and ABTS radical scavenging assays.The results indicated that the optimal extraction process for Ginkgo biloba L.leaf polysaccharides using ammonium oxalate was identified as follows:extraction temperature of 95℃,extraction time of 1 hour and 45 minutes,ammonium oxalate concentration of 1.00%,and liquid to material ratio of 16 mL/g.Under these conditions,the extraction yield of Ginkgo biloba L.leaf polysaccharides was(8.26±0.69)%,close to the predicted value.The ammonium oxalate-extracted polysaccharides,at a concentration of 5 mg/mL,demonstrated significant radical scavenging activity,achieving 88.18%and 86.27%scavenging rates for DPPH and ABTS+radicals,respectively.This activity was comparable to that of the positive control.These findings indicated that ammonium oxalate extraction was an effective method for obtaining Ginkgo biloba L.polysaccharides with high yield and potent antioxidant properties.关键词
